[ixpmanager] E-mail environment

First question, do edits to application.ini take effect immediately? 

Original question, what's the e-mail environment like? I see that I'm specifying e-mail addresses throughout the config, but there's only one request for a username\password. Does all mail just send as that host, username, password? What all e-mail addresses in the config need to be able to receive mail?
